10 Evidence-Based Integrative Therapies That May Enhance 5-FU Chemotherapy in Colorectal Cancer. Several evidence-based non-conventional therapies have been studied for their ability to enhance the anti-cancer effects of 5-fluorouracil (5-FU) in colorectal cancer. While most are not considered standard of care, many have preclinical evidence, and some have early clinical data suggesting synergy with 5-FU.

My name is Bernie Davis. I was recently diagnosed with colon cancer. See my diagnostic info below. As you can read from the discussion about my stage at diagnosis, while the percentage of risk of relapse is debatable, I do have a risk of relapse. A level of risk that I consider unacceptable.

Therefore, I have chosen to work with PeopleBeatingCancer.org to undergo integrative therapies to reduce my risk of relapse as well as reduce my risk of side effects.

In addition to the 10 natural compounds shown to reduce my risk of colon cancer, I also supplemented with Fenbendazole.

As of June of ’26, I have completed 12 rounds of Folfox chemotherapy while also undergoing the evidence-based, non-conventional therapies listed below.

The side effects I’ve experienced over the past six months are manageable. I’ve recently experienced real fatigue, but I think it’s less than what I read about other colon cancer patients experience.

I can’t report any long-term side effects. Nor can I report anything about a relapse. No news is good news at this point 😁

Enhance 5-FU Chemotherapy in Colorectal Cancer



✅ Summary in plain terms:
The report describes a 3.1 cm adenocarcinoma that has grown all the way through the intestinal wall to the abdominal lining, shows signs of spreading through blood vessels, lymph channels, and nerves, and has spread to 7 of 24 lymph nodes plus one additional tumor deposit. Fortunately, the surgeon removed it completely with negative margins. The official pathologic stage is pT4a pN2b (Stage IIIc).


Overall survival and recurrence statistics for Stage IIIC

  • In older studies, when Stage III is subdivided by both T and N, Stage IIIC is the worst prognostic subgroup within Stage III. One classic analysis (ERCRC) estimated 5-year survival for Stage IIIC to be around 30 %(versus ~80 % for IIIA, ~60 % for IIIB) in patients treated with surgery alone. ACS Journals
  • More recent population-level data suggest somewhat more favorable outcomes in the modern era, with intensification of therapy and better supportive care. PMC+1
  • For example, the SEER “regional disease” category (which includes Stage III) reports a 5-year relative survival of ~73 %. American Cancer Society
  • However, that “regional” group pools all Stage II and III—so the survival specifically for Stage IIIC is lower than the average for “regional” disease.

So, in a patient with Stage IIIC colon cancer and good margins, one might approximate a 5-year overall survival in the ballpark of 40-60 % (this is a rough, optimistic estimate in a favorable scenario). In more aggressive disease or with less optimal patient factors, it may be lower.

Enhance 5-FU Chemotherapy in Colorectal Cancer


1. Curcumin (Strongest Evidence)

Curcumin is one of the most extensively studied natural compounds in colorectal cancer.

Evidence

  • Curcumin appears to sensitize colorectal cancer cells to 5-FU.
  • May inhibit pathways associated with chemotherapy resistance (NF-κB, STAT3, Wnt/β-catenin).
  • Several studies have demonstrated increased apoptosis and reduced tumor growth when combined with 5-FU.

Human Research

The phase IIa CUFOX trial combined curcumin with FOLFOX chemotherapy in metastatic colorectal cancer and found the combination was safe and showed encouraging survival outcomes.

2. Vitamin D

Vitamin D deficiency is common among colorectal cancer patients.

Evidence

  • Higher vitamin D levels correlate with improved colorectal cancer survival.
  • Laboratory studies suggest vitamin D enhances 5-FU sensitivity.
  • May regulate tumor cell differentiation and immune function.

Clinical Data

The SUNSHINE trial found high-dose vitamin D supplementation improved progression-free survival in metastatic colorectal cancer patients receiving chemotherapy.

3. Omega-3 Fatty Acids (EPA/DHA)

Evidence

Omega-3 fatty acids may:

  • Increase chemotherapy sensitivity
  • Reduce inflammation
  • Reduce cachexia and treatment-related weight loss

Research Findings

EPA appears to:

  • Enhance apoptosis induced by 5-FU
  • Improve drug uptake into tumor cells
  • Reduce chemotherapy resistance mechanisms

4. Melatonin

Melatonin is far more than a sleep supplement.

Evidence

Research demonstrates that melatonin:

  • Enhances 5-FU-induced apoptosis
  • Protects normal tissues from oxidative damage
  • May reduce chemotherapy side effects

Potential Mechanisms

  • Antioxidant activity
  • Immune modulation
  • Mitochondrial regulation
  • Suppression of angiogenesis

Several integrative oncology centers routinely include melatonin in supportive care protocols.


5. Medicinal Mushrooms

Particularly:

  • Turkey Tail
  • Maitake
  • Reishi

Evidence

These mushrooms:

  • Enhance NK cell activity
  • Improve immune surveillance
  • May help patients tolerate chemotherapy better

While direct synergy with 5-FU is less established than curcumin, they may improve host immune response during treatment.

6. Probiotics and Gut Microbiome Therapy

Emerging evidence suggests the gut microbiome influences chemotherapy response.

Evidence

Specific probiotic strains may:

  • Reduce chemotherapy-induced diarrhea
  • Improve gut barrier function
  • Enhance anti-tumor immunity

Some studies suggest certain bacterial species increase responsiveness to fluoropyrimidine-based chemotherapy.


7. Green Tea Polyphenols (EGCG)

Evidence

EGCG has demonstrated:

  • Enhanced 5-FU cytotoxicity

Mechanisms

8. Exercise

Some research suggests exercise-induced changes in tumor blood flow may improve chemotherapy delivery.


9. Modified Citrus Pectin

Evidence

Modified citrus pectin inhibits galectin-3, a protein involved in:

  • Tumor growth
  • Metastasis
  • Chemotherapy resistance

Preclinical studies suggest potential synergy with conventional therapies, though colorectal cancer-specific clinical evidence remains limited.


10. Fasting-Mimicking Diets (FMD)

Evidence

Research from Valter Longo and colleagues suggests fasting or fasting-mimicking diets may:

  • Increase cancer cell sensitivity to chemotherapy
  • Protect healthy cells from chemotherapy damage

Early human studies are promising but remain investigational.

Therapies Requiring Caution

These should not be used without oncology supervision:

  • High-dose antioxidants during active chemotherapy
  • High-dose vitamin C administered around infusion days
  • Herbal mixtures with unknown ingredients
  • Fenbendazole, ivermectin, or mebendazole during chemotherapy unless supervised by an oncologist familiar with potential interactions
